Three Compounds, Three Different Risk Profiles

How a copper peptide, an untested three-peptide blend, and a melanocortin agonist differ in mechanism, evidence, and what is actually known.

The short version

GHK-Cu has the most human data and the only approved use, as a topical cosmetic ingredient. GLOW combines GHK-Cu with two other peptides but has never been tested as a blend. Melanotan II has old, small human trials and a long, serious case-report record.

Reading them together makes the differences clear: more evidence does not mean fewer safety concerns, and less regulation does not mean less mechanism. Each compound's profile stands on its own.

Mechanism

The three compounds act through unrelated biological pathways. GHK-Cu is a copper carrier and matrix-remodeling signal: at very low concentrations it drives skin fibroblasts to build collagen, elastin, and connective-tissue matrix, and its copper ion enables the cross-linking that makes new tissue stable [1][4][7]. GLOW layers GHK-Cu's matrix effect together with BPC-157's blood-vessel-growth and tissue-protective activity and TB-500's cell-migration activity — three separate mechanisms combined in one product, never in one study [8][10]. Melanotan II is structurally and functionally unrelated to either: it is a non-selective melanocortin receptor agonist that switches on pigment production in skin cells and appetite/arousal circuits in the brain, through a cAMP-driven signaling cascade [15]. Only GHK-Cu, and the GHK-Cu portion of GLOW, touch skin matrix biology directly; Melanotan II's route to 'aesthetics' runs entirely through pigment, not structure.

Evidence base

GHK-Cu carries the most complete human evidence of the three: a six-month, 45-person controlled hair-growth trial [3], decades of collagen-synthesis and wound-healing literature [4][6][7], and a 2025 review quantifying its anti-wrinkle efficacy against established comparators [1]. Much of that foundational work traces to a small number of research groups, which limits independent replication but does not erase the trial data.

GLOW has the thinnest direct evidence: no trial has tested the three-peptide combination at any dose. Every claim rests on BPC-157 [9][10], TB-500, or GHK-Cu [4][6] studied alone — a 2026 review is the only source that discusses all three peptides together, and even it evaluates them individually, not as a formulated blend [8].

Melanotan II has the oldest human trial data — a 1998 crossover study in 10 men [15] — but almost nothing since in controlled conditions. What has accumulated instead is a case-report literature: documented instances of renal injury, priapism, and melanoma in people who used it outside any trial [13].

Regulatory status

Regulatory standing tracks roughly with mechanism, not with how the products are marketed. Topical GHK-Cu — Copper Tripeptide-1 — is a legal cosmetic ingredient in the US, EU, and UK; injecting it or taking it systemically has no approved pathway anywhere. GLOW's components split three ways: GHK-Cu's topical form is a cosmetic ingredient, while BPC-157 and TB-500 are unapproved research chemicals with no therapeutic approval, and TB-500 (as the synthetic thymosin beta-4 fragment) is named on the World Anti-Doping Agency's Prohibited List at all times [8]. Melanotan II has never been approved by any regulator for any indication — no completed Phase II or III trial exists, and regulators in multiple countries have issued specific warnings against melanotan-containing tanning products [14].

Key caution

Each compound carries a defining caveat that should not be averaged away. For GHK-Cu, it is that the well-documented safety and efficacy record is topical-only — systemic and injectable use is unstudied, and repeated systemic copper exposure carries a theoretical accumulation concern. For GLOW, it is that the blend itself has no safety data: three peptides with different clearance rates, combined in one product, and one of them (TB-500) carries an anti-doping prohibition that has nothing to do with skin [8]. For Melanotan II, it is the growing case-report record of serious harm — new and changing moles, melanoma, renal infarction, and priapism — set against a product that was never approved and is sold without quality control [13]. Reading the three together, evidence volume and safety risk move independently, not together.
